I used Waves Multirack which is a piece of software for running multiple instances of plugins on multiple channels live, and configured 4 different reverb/delay plugins. I had a 4i/8o interface that I configured as 4 mono FX inputs and 4 stereo FX returns into the desk. I got started running plugins live by treating my laptop like a really awesome FX box. This is usually only done on huge tours with lots of money for redundancy. From a laptop, you can remotely control the plugins, and having a network of DSP servers means that a number of the servers can fail during a show but the remaining servers pick up the slack. Basically, you've got a fancy console with a few awesome computers connected to it. The step up from running your FX on a laptop is getting a dedicated DSP server (or servers). If you're using a digital console, you'll probably want an interface that speaks MADI as that's pretty common.

If you're using an analog console, your interface needs a bunch of analog line level i/o and then you can patch the interfaces's i/o over the console's insert points or groups or however. You can bring a laptop and a means to interface your laptop into the audio. Most desks do not have the ability to run plugins natively within the console, so you need to add some form of external processing.

There are engineers that tour with a thumb drive that contains the installers for all their plugins, and an iLok which authorizes the desk to use em. Some desks, most commonly the AVID SC48 and the PROFILE can run waves plugins using the processing power of the desk.
